Endogenous miRNA has attracted a lot attention because of its critical role in the regulation of gene expression during embryonic development. The design of shRNA transcripts, as primary microRNA (miR) mimics (miR-shRNAs) driven by Pol II promoters which significantly expands the possibilities for conditional RNAi in mammalian cells. This technology minimizes the problems caused by long-term gene suppression from Pol II-shRNA; implying an ideal application for gene therapy with a controllable gene knockdown.
